Delay discounting in youth at clinical high-risk for psychosis and adults with schizophrenia

Background
Schizophrenia (SZ) is typically preceded by a prodromal (i.e. pre-illness) period characterized by attenuated positive symptoms and declining functional outcome. Negative symptoms are prominent among individuals at clinical high-risk (CHR) for psychosis (i.e. those with prodromal syndromes) and highly predictive of conversion to illness. Mechanisms underlying negative symptoms in the CHR population are unclear. Two studies were conducted to evaluate whether abnormalities in a reward processing mechanism thought to be core to negative symptoms in SZ, value representation, also exist in CHR individuals and whether they are associated with negative symptoms transphasically.
MethodsStudy 1 included 33 individuals in the chronic phase of illness who have been diagnosed with schizophrenia or schizoaffective disorder (SZ) and 40 healthy controls (CN). Study 2 included 37 CHR participants and 45 CN. In both studies, participants completed the delay discounting (DD) task as a measure of value representation and the Brief Negative Symptom Scale was rated to measure negative symptoms.
ResultsResults indicated that patients with SZ had steeper discounting rates than CN, indicating impairments in value representation. However, CHR participants were unimpaired on the DD task. In both studies, steeper discounting was associated with greater severity of negative symptoms.
ConclusionsThese findings suggest that deficits in value representation are associated with negative symptoms transphasically.

We present a detailed overview of the cosmological surveys that we aim to carry out with Phase 1 of the Square Kilometre Array (SKA1) and the science that they will enable. We highlight three main surveys: a medium-deep continuum weak lensing and low-redshift spectroscopic HI galaxy survey over 5 000 deg2; a wide and deep continuum galaxy and HI intensity mapping (IM) survey over 20 000 deg2 from $z = 0.35$ to 3; and a deep, high-redshift HI IM survey over 100 deg2 from $z = 3$ to 6. Taken together, these surveys will achieve an array of important scientific goals: measuring the equation of state of dark energy out to $z \sim 3$ with percent-level precision measurements of the cosmic expansion rate; constraining possible deviations from General Relativity on cosmological scales by measuring the growth rate of structure through multiple independent methods; mapping the structure of the Universe on the largest accessible scales, thus constraining fundamental properties such as isotropy, homogeneity, and non-Gaussianity; and measuring the HI density and bias out to $z = 6$ . These surveys will also provide highly complementary clustering and weak lensing measurements that have independent systematic uncertainties to those of optical and near-infrared (NIR) surveys like Euclid, LSST, and WFIRST leading to a multitude of synergies that can improve constraints significantly beyond what optical or radio surveys can achieve on their own. This document, the 2018 Red Book, provides reference technical specifications, cosmological parameter forecasts, and an overview of relevant systematic effects for the three key surveys and will be regularly updated by the Cosmology Science Working Group in the run up to start of operations and the Key Science Programme of SKA1.

Orlistat accentuates the fat-induced fall in blood pressure in older adults

Postprandial hypotension may be influenced by the digestion of fat. The aim of the present study was to evaluate the hypothesis that products of fat digestion mediate the hypotensive response to fat. In part A of the study, nine healthy older subjects were studied on three separate occasions in randomised order. Blood pressure, heart rate (HR), plasma TAG and gastric emptying were measured following the ingestion of equivolaemic drinks: (1) 300 ml of high-fat drink (88 % fat); (2) fat drink mixed with 120 mg orlistat (lipase inhibitor); (3) water (control). In part B of the study, ten healthy older subjects were studied on two separate occasions. Blood pressure, HR, plasma TAG and superior mesenteric artery flow were measured during 90 min intraduodenal infusions of 10 % intralipid (2·7 ml/min), with and without 120 mg orlistat. Oral fat ingestion was associated with decreases in systolic and diastolic blood pressures (both P = 0·0001) that were greater when orlistat was co-administered (both P < 0·05), and an increase in HR (P = 0·0001) that was inhibited by orlistat co-administration (P < 0·03). Gastric emptying was slowed by oral fat digestion, and orlistat administration inhibited this slowing (P < 0·04). Intraduodenal fat infusion was not associated with changes in blood pressure but increased HR (P < 0·0001), an effect attenuated by orlistat (P < 0·05). In conclusion, orlistat potentiates the hypotensive response to oral fat in older adults, possibly as a result of faster gastric emptying of fat. The results do not support a role for fat digestion in lowering blood pressure.

We present results of the Australia Telescope Compact Array (ATCA) radio continuum observations of massive stars in the Sco OB1 association. Most stars detected in the program show spectral indices lower than value expected from thermal free-free emission.

Introduction and overview
Growth hormone (GH) releasing peptides (GHRPs) were developed by Bowers and co-workers in the early 1980s, specifically as GH secretagogues (Bowers et al., 1980; Bowers et al., 1991). These small, synthetic peptide molecules, created by modifying the structure of met-enkephalin, include the hexapeptides GHRP-6 (His-D-Tryp-Ala-Trp-D-Phe-Lys-NH2), -1, -2 and hexarelin (His-D-2–Methyl-Try-Ala- Trp-DPhe-Lys-NH2), in which the D-tryptophan has been substituted with its 2-methyl derivative. Oral administration of the GHRPs stimulates GH release but the bioavailability, and hence GH stimulatory potency, is much greater after parenteral administration; an oral GHRP-6 dose of 300 µg/kg produces a comparable increase in serum GH concentration to a 1 µg/kg intravenous (iv) bolus (Bowers et al., 1992; Hartman et al., 1992). The onset of action of GHRP is somewhat slower after oral than iv administration, however the duration of action is similar for both, averaging about 120–150 minutes (Hartman et al., 1992). The reduced potency of oral GHRP and this relatively limited duration of action prompted the search for GH secretagogues with a longer duration of action and greater oral bioavailability (see below).
Preliminary data indicate that it may be possible to administer GHRP transdermally (Fleisher et al., 1995), and intranasal administration of hexarelin has been shown to stimulate GH secretion in healthy elderly people and increase growth velocity and serum insulin-like growth factor-I (IGF-I) concentrations in short children (Laron et al., 1995). The GHRPs are well tolerated and to date few side effects have been reported. The acute stimulatory effects of GHRP-6 on GH secretion are independent of gender (Penalva et al., 1993a) and there are no reports of gender-related differences in GH responses to the other GHRPs.

Experimental and theoretical data are used to compare the effect of three possible leach mechanisms for borosilicate glass waste buried in a granite host-rock on the release and subsequent migration of 135Cs. Protracted release episodes and variations of up to an order of magnitude in groundwater transport times and five orders in output concentrations are possible.

Much effort is currently centred on the construction of elaborate computer programs to model the release to the biosphere of radionuclides from proposed high level radioactive waste repositories in deep geological formations. Although the lack or poor quality of background data is often emphasised, it is generally considered that by examining a wide range of possible values of the important parameters involved, likely processes are assessed or their omission and possible relevence is acknowledged.
